Previous studies using reverse transcriptase PCR have shown increased nAChR receptor subunit expression with nicotine treatment. We proposed to treat the H69 human lung carcinoma cell line with nicotine and observe the effects on gene expression using real-time PCR with the hypothesis that nicotine treatment alters gene expression of alpha4-alpha2 subunits. In analyzing a 4 day 100 M nicotine treatment of the H69 cell line, a significant increase in gene expression of the alpha4 subunit was observed after 24 hours.
