PCV1 is a non-enveloped, non-virulent, single-stranded DNA virus that infects pigs. Two related viruses, CAV and PCV2, encode proteins capable of selective induction of apoptosis in transformed cells, which is associated with cell-type specific localization. Our project is the first to examine the expression and localization of PCV1 VP3. Sequence analysis suggests NES and NLS motfis may exist in the protein. Expression of GFP-tagged PCV1 VP3 in H1299 cells localized to the cytoplasm, suggesting a functional NES may be present. The observed decrease in confluency of H1299 cells following transfection with the p3XFLAG-myc-CMV-26-PCV1 ORF3 vector suggests PCV1 VP3 may be capable of inducing apoptosis in transformed cells.
